چکیده

Badakhsh, Fred F. (University of Georgia, Athens), and John W. Foster. Detoxification and immunogenic properties of endotoxin-containing precipitate of Brucella abortus. J. Bacteriol. 91:494-498. 1966.-Endotoxin-containing precipitates (ECP) were prepared from Brucella abortus strain 19A by aqueous ether extraction followed by ethyl alcohol precipitation. Lysozyme was the most effective of several enzymes tried for detoxification of endotoxin present in the precipitate. Trypsin was shown to reduce mouse lethal toxicity but not rabbit dermal toxicity. Immunological studies of ECP and enzyme-treated ECP demonstrated that lysozyme did not harm the immunogenic property of ECP, whereas heat, ribonuclease, lipase, and proteolytic enzymes had an adverse effect. Serological reactivity of ECP was increased after lysozyme treatment, whereas ribonuclease reduced serological activity.

Immunogens of Brucella Abortus S19 Identified By Two-Dimensional Gel Electrophoresis and Immunoblotting

Background: Lipopolysaccharides (LPSs) and several antigenic proteins of Brucella have been considered for preparation of diagnostic reagents and subunit vaccines. The objective of this study was to identify and compare immunogens of B. abortus S19 which induce humoral immune responses in human, goat and rabbit. Material and Methods: The bacterial whole cell extract was prepared in extraction b...

Some structural and biological properties of Brucella endotoxin.

Hot phenol-water extraction of smooth Brucella abortus and B. melitensis cells yielded a toxic fraction which was recovered from the phenol phase (fraction 5). Chemically, fractions 5 from both Brucella species were lipid-carbohydrate-protein-2 keto-3-deoxyoctulosonic acid complexes which were stable to heat and resistant to Pronase digestion. Electron micrographs of the Brucella toxins were mo...

High Level Expression of Recombinant Ribosomal Protein (L7/L12) from Brucella abortus and Its Reaction with Infected Human Sera

Brucellosis, caused by Brucella spp., is an important zoonotic disease that causes abortion and infertility in cattle and undulant fever in humans. Various studies have examined cell-free native and recombinant proteins as candidate protective antigens in animal models. Among Brucella immunogenes, antigen based on ribosomal preparation has been widely investigated. In this study, the immunogeni...

Induced sensitization of normal laboratory animals to Brucella abortus endotoxin.

A Boivin preparation of Brucella abortus, unlike common enterobacterial endotoxins, failed to depress water intake or increase numbers of hemolysin-producing spleen cells in mice, or to cause delayed inflammatory reactions in rabbit skin. Reactivity to the B. abortus endotoxin was found only in animals which were previously given the endotoxin with, but not necessarily in, complete Freund's adj...
